摘  要:目的通过建立糖尿病大鼠模型,探讨糖尿病周围神经痛大鼠脊髓背角JNK的水平。方法雄性SD大鼠腹腔注射链尿佐菌素(STZ)诱导大鼠糖尿病变模型,随机分为空白对照组(C组)、糖尿病对照组(DC组)和糖尿病周围神经痛组(D组)(n=6)。各组分别在注射前、注射后4、6及8周后测定机械缩足反射阈值(MWT)并行脊髓背角p-JNK免疫组织化学检测。结果 D组及DC组大鼠注射STZ后4周机械痛敏形成,并维持至本实验观察时间终点注射STZ后8周。与注射STZ前及C组大鼠MWT比较,差异均有统计学意义(P<0.05)。D组大鼠注射STZ4周时,脊髓背角磷酸化JNK免疫反应阳性细胞数增加,注射STZ后6周及8周,脊髓背角磷酸化JNK免疫反应阳性细胞数增加增加明显,与注射STZ前及C组大鼠比较差异均有统计学意义(P<00.05)。结论糖尿病周围神经痛大鼠脊髓背角JNK的激活,可能参与糖尿病周围神经痛的发生与发展。【Objective】To investigate the changes in JNK expression in spinal dorsal horn in a rat model of painful diabetic peripheral neuropathy induced by intraperitoneal injection of streptozotocin.【Methods】Male SD rats were randomly divided into 3 groups(n =6 each):sham operation group(sham),diabetic control group(DC group) and diabetic group(D group).Mechanical withdrawal threshold was measured in rats in each group before(baseline) intraperitoneal injection and at 4w,6w and 8w after injection.Rats in each group were killed to remove L5 spinal for determination of p-JNK at a counterpart time respectively.【Results】WT of rats in D group and DC group was reduced 4w after intraperitoneal injection and maintain till the end of this study compared to the base line and sham injection group.p-JNK expression in spinal dorsal horn of rats in D group was up-regulated 4w after intraperitoneal injection and maintain this level till the end of this study compared to the baseline and sham group,DC group.【Conclusion】Up-regulation of JNK expression in spinal dorsal horn is involved in the mechanism of painful diabetic peripheral neuropathy.
