摘  要:对采自黑龙江省某动物园的东北虎源球虫虫种进行鉴定。对东北虎源球虫进行形态学观察;应用孢子化卵囊对犬进行感染性试验;根据已发表的猫等孢球虫核糖体内转录间隔区Ⅰ(ITSⅠ)序列设计引物,提取虫体基因组DNA扩增目的片段,并与其他球虫的ITSⅠ序列进行同源性比较。结果表明,根据形态学观察发现该球虫符合等孢属球虫的特征;用孢子化卵囊感染犬,结果不能在其粪便中检出卵囊;利用聚合酶链式反应(PCR)扩增的目的片段,与其他19种球虫的ITSⅠ序列同源性为35.97%~98.71%之间。通过对东北虎源球虫形态学观察、犬的感染性试验与分子鉴定,确定东北虎源球虫为等孢属球虫,并且与猫等孢球虫亲缘最近。The purpose of the paper was to identify coccidian the Siberian tigers from a zoo in Heilongjiang Province. The morphology observation of coccidian oocysts was carried out. The dogs were infected with the coccidian oocysts. The specific PCR primerwas designed from Cystoisospora felis internal transcribed spacerⅠ(ITSⅠ). The target fraction was amplified by PCR and the homology was analyzed with the othercoccidian. The results showed that the morphology observation of coccidian oocysts of the Siberian tigershowed that the morphous was conformity to the feature of Isospora. Oocysts could not be detected in feces of dogs inoculated with sporuled oocysts. The target fraction of 389 bp was amplified and the homology among other 19 coccidians was from 35.97% to 98.71%. According to the morphology observation, the infection experiment on dogs and the comparison of ITSⅠfractions, it could be confirmed that the coccidian oocystfrom Siberian tigers was Isospora which was closerto Isospora felis than others.
