四川‘竹根姜’试管种苗产业化生产的技术因素分析

摘  要:将姜茎尖接种于不同激素的培养基中诱导丛生芽,在含有不同铵盐浓度、不同激素浓度的培养基上增殖和诱导生根以及在不同配比的基质中移栽试管苗,结果表明:MS+6-BA2.0mg/L+NAA0.1mg/L浓度配比为最佳诱导培养基;以含1/3NH4NO3的MS培养基为基本培养基,6-BA1.5mg/L+NAA0.1mg/L配比时,增殖系数达到2.43且玻璃化率最低;6-BA0.5~1.0mg/L+NAA0.2mg/L的激素浓度配比最适合四川‘竹根姜’生根,芽苗生根率达100%,根系健壮且有侧芽萌发;珍珠岩∶泥炭土体积比为3∶7的移栽成活率最高达96.67%,且芽苗健壮,生长快.The important technology factors for plantlet industrialized production in ‘Zhugenjiang’ ginger (Zingiber officinale Rosc.) via tissue culture were studied. Shoot-tips of ginger were inoculated onto MS medium containing different plant hormones to induce adventitious buds,the induced plantlets were then cultured on proliferation and rooting media and,finally,the rooted plantlets were transferred to different substrates. The suitable shoot-inducing medium was MS+6-BA 2.0 mg/L+NAA 0.1 mg/L. On MS (containing 1/3 NH4NO3)+6-BA 1.5 mg/L+NAA 0.1 mg/L,a propagation ratio of 2.43 was obtained with the lowest vitrification rate. The most suitable rooting medium was MS+6-BA 0.5~1.0 mg/L+NAA 0.2 mg/L. Under this condition,the rooting ratio reached 100% and the roots grew well and produced adventitious buds. The optimum substrate combination was 3∶7 of pearlite∶peat soil (v/v),on which the survival rate of ginger was 96.67% and the transplanted plants grew vigorously.
