用红外光谱研究胃癌细胞SGC7901光敏损伤后蛋白质的二级结构

摘　　要：本文用傅立叶变换红外光谱研究光动力作用胃癌细胞SGC7901后,蛋白质二级结构的变化。对蛋白质的酰胺I带,采用去卷积,二阶导数谱进行曲线拟合,得到蛋白质二级结构的峰个数和位置以及各二级结构的含量,结果显示蛋白质无规卷曲和β转角的含量上升,α螺旋、β折叠含量减少,蛋白质空间构象改变。实验结果表明,光动力对胃癌细胞SGC7901蛋白质二级结构造成明显损伤。The paper is intended to study the change of the secondary structure of proteins stomach cancer cell SGC7901 after photodynamic therapy by FTIR.For the amide I band of protein,we obtain both the secondary structure of proteins peak number and peak position of the secondary structure of proteins and contents of the secondary structure of proteins by deconvolved second-derivative spectra curve-fitting.Results are showed that the content of random coils and β turns increase dramatically and the content of α helixes and β sheets decrease and the protein conformation alter at same time.It is prompted that there are obvious damages for the secondary structure of proteins of stomach cancer cell SGC7901 after photodynamic therapy.
