摘  要:目的研究乌灵菌粉对大鼠戊四唑点燃癫痫形成过程的作用和慢性癫痫诱发记忆障碍的作用及机制。方法将SD大鼠随机分为4组:戊四唑点燃模型组(模型对照组),低剂量乌灵菌粉(0.3 g.kg-1,ig)组,高剂量乌灵菌粉(0.6 g.kg-1,ig)组和空白对照组。各组大鼠隔日腹腔注射亚惊厥剂量(35 mg.kg-1)的戊四唑(空白对照组注射生理盐水),共12次。乌灵菌粉在每次注射戊四唑前0.5 h灌胃。采用穿梭箱被动回避试验测定大鼠的记忆能力,化学荧光法测定脑内组胺含量。结果乌灵菌粉能延缓大鼠戊四唑点燃癫痫的形成过程,但不能阻断点燃的最终形成。戊四唑点燃癫痫使大鼠在穿梭箱中的被动回避时间缩短,乌灵菌粉明显逆转了这种被动回避时间的缩短。戊四唑点燃癫痫使大鼠脑内组胺含量明显下降,乌灵菌粉部分逆转了这种下降,在海马和下丘脑达到了统计学显著性意义。结论乌灵菌粉能延缓大鼠戊四唑点燃癫痫的形成过程,改善癫痫诱发的记忆障碍,其作用可能与增强脑内组胺能神经的功能有关。OBJECTIVE To investigate the effects of Wuling mycelia on seizure development and memory impairment induced by pentylenetetrazole-kindling epilepsy in rats.METHODS SD rats were randomly divided into four groups: pentylenetetrazole-kindling model group,low dose of Wuling mycelia(0.3 g·kg^-1,ig) group,high dose of Wuling mycelia(0.6 g·kg^-1,ig) group and control group.The rats were intraperitoneal injected with a subconvulsive dose(35 mg·kg^-1) of pentylenetetrazole(saline in control group) every 48 h for 12 times.Wuling mycelia were intragastrically applied 30 min before every pentylenetetrazole injection.Memory was evaluated by shuttle box with passive avoidance and brain histamine was measured.RESULTS Wuling mycelia the kindling induced by pentylenetetrazole.The latency of passive avoidance of pentylenetetrazole-kindling model group was significantly shortened in shuttle box,which was reversed by Wuling mycelia.Brain histamine contents decreased after kindling induced by pentylenetetrazole,which were partially reversed by Wuling mycelia,especially in hippocampus and hypothalamus.CONCLUSION Wuling mycelia could postpone the kindling and ameliorate memory impairment induced by pentylenetetrazole kindling.The enhancement of neuronal histamine activity was one of possible mechanisms.
