摘 要:目的:建立以反相高效液相色谱法测定人血清中拉莫三嗪浓度的方法。方法:色谱柱为KromasilC18,流动相为甲醇∶乙腈∶0.03mo·lL-1醋酸铵(乙酸调pH值至3.5±0.1)=12∶12∶27;流速为1.0mL·min-1,柱温为室温,检测波长为310nm,进样量为20μL。结果:拉莫三嗪血药浓度在0.5~30.0mg·L-1范围内线性关系良好(r=0.9986);方法回收率为99.00%~102.00%;日内、日间RSD分别为1.66%~2.50%、2.18%~2.98%。结论:本方法操作简便、快速、灵敏、重复性好、回收率及精密度高,可用于拉莫三嗪的治疗药物监测。OBJECTIVE:To develop an RP-HPLC method for the content determination of lamotrigine in human serum.METHODS:HPLC method was conducted on Kromasil C18(250 mm×4.6 mm,5 μm)column with the mobile phase consisted of methanol-acetonitrile-0.03 mol·L-1 NH4AC (pH value adjusted to 3.5±0.1 with acetic acid)=12:12:27.The flow rate was set at 1.0 mL·min-1 and the detection wavelength were at 310 nm.The column temperature was room temperature and injection volume was 20 μL.RESULTS:The linear range of lamotrigine were 0.5~30.0 mg·L-1 (r=0.998 6) with recovery of 99.00%~102.00%.The RSD of intra-day and inter-day were 1.66%~2.50%,2.18%~2.98% respectively.CONCLUSION:The method is simple,rapid,sensitive and reproducible with high recovery and good precision for pharmaceutical monitoring of lamotrigine.
