摘 要:用Fenton反应产生羟基自由基(OH·),用电子自旋捕集技术(EPR)测定了四甲基哌啶氮氧自由基及衍生物(4-TEMPO)对OH·的捕获能力。用吩嗪硫酸二甲酯(PMS)-NADH体系产生超氧阴离子自由基(O—·2),用NBT还原法比较了4-TEMPO对O—·2的清除能力。结果显示:①两个四甲基羟哌啶(HTMPOH、OTMPOH)和两个四甲基哌啶氮氧自由基(HTMPO、OTMPO)在40~160μmol·L-1能清除O—·2。②4-TEMPO在抗氧化浓度甚至低于此浓度均使DMPO-OH·(二甲基吡咯啉氮氧化合物与OH·形成的加合物)的信号明显高于对照。两个阴性对照四甲基哌啶(HTMP。The scavenging activities for O · 2 and OH· of 4 oxy and 4 hydroxy Nitroxides and its derivatives(4 TEMPO)were studied.Superoxide anions(O · 2)generated in a phenazin methosulphate(PMS) NADH system were assayed by the reduction of mitroblue tetrazolium(NBT).Hydroxyl radicals(OH)·generated in “Fenton reaction” were detected by spin trapping technique.Nitroxide free radicals,HTMPO、OTMPO and 1 hydroxyl compounds HTMPOH、OTMPOH can scavenging O · 2 during the concentrations of 4 TEMPO are 40 160μmol·L -1 .The effect is clearly different from the control group(P<0 05 or P<0 01)·HTMP、HTMPO and 1 hydroxyl compounds can t reduce the signal of DMPO OH· during their concentrations are 1 6 160μmol·L -1 ,but the signal of DMPO OH·is raise.HTMP and OTMP have no effect for OH·and O · 2.Which indicated that the mechnnism of scavenging O · 2 may be related to the stopping radical chain reation directly or analogy with superoxide dismutase(SOD).It is necessary that the mechanisms of effect for OH·or DMPO of 4 TEMPO were studied further.
