茸毛链球菌主要表面蛋白抗原的定性分析

摘　　要：目的：提取茸毛链球菌主要表面蛋白抗原（ＰＡｇ）对其性质进行初步鉴定。方法：用硫酸铵沉淀、ＤＥＡＥ－ｃｅｌ－ｌｕｌｏｓｅ离子交换和 Ｓｅｐｈａｒｏｓｅ ６Ｂ凝胶过滤从茸毛链球菌 ６７１５培养上清中提取 ＰＡｇ，并对其分子量，蛋白质含量，氨基酸组成及免疫学特性进行检测。结果与结论：ＰＡｇ能与特异性抗血清发生反应，ＳＤＳ－ＰＡＧＥ呈单一区带，分子量为２１０ＫＤ，蛋白质含量为６１．４％，氨基酸组成分析揭示其酸性和碱性氨基酸分别为２６．７％和１０．７％。Objective: To purify and characterize the major cell surface protein antigen of streptococcus sobrinus. Methods: PAg was purified by 65 % saturate ammonium sulfate precipitation, DEAE - cellulose ion - exchange chromatography and Sepharose 6B gel - filtration chromatography. Results and Conclusion: SDS-PAGE showed the extract PAg was pure and in the ranges of 210KD, protein quantitative analysis showed PAg contained 61.4 % proteins, amino acid analysis revealed that PAg contained 26. 7 % acidic and 10.7 % basic acid residues.
