摘 要:目的:探讨小鼠肝内胆红素葡萄糖醛酸化的性别差异及原因。方法:提取C57BL/6J小鼠肝微粒体,检测胆红素尿苷二磷酸葡萄糖醛酸转移酶(BUGT)的活性及LubrolPx激活后活性,并以Northern印迹杂交检测肝内BUGT基因的表达,比较雌、雄小鼠之间的差别。结果:雄性小鼠BUGT活性比雌性小鼠高,激活后BUGT活性雄性小鼠比雌性小鼠更高,雄性小鼠肝内BUGT基因有更高mRNA水平的表达。Objective: To study the gender related differences in glucuronidation of bilirubin and its cause. Methods: Microsomes were extracted from livers of C57BL/6J mice, the activities of both native and activated bilirubin UDP glucuronosyltransferase (B UGT) with Lubrol Px in microsomes of mice were measured, and the expression of hepatic B UGT gene in both male and female mice was analysed by Northern blotting. Results: The male mice not only had higher enzymatic activities in both native and activated B UGT than that of the female mice, but also exhibited stronger expression level of hepatic B UGT gene mRNA in male mice than that in female mice. Conclusion:A significant gender related difference did exist in the glucuronidation of bilirubin in mice. The stronger expression level of hepatic B UGT gene mRNA in male mice might be responsible for the above mentioned activity differences.
