摘  要:研究海洋放线菌WB-F5发酵液的抑菌活性,并分离纯化其抑菌作用物。通过琼脂打孔法测定WB-F5发酵液对各菌株的抑制活性。本实验选用大孔树脂吸附法、硅胶柱层析法等方法,分离纯化活性物质。WB-F5发酵液对金黄色葡萄球菌、耐甲氧西林金黄色葡萄球菌(MRSA)、枯草杆菌等革兰氏阳性菌有较好的抑制作用,同时确定大孔吸附树脂AB-8和硅胶柱层析作为分离手段较为适宜,并且得到化合物WBF5-F,经结构解析证明其为5,7,4′-三羟基异黄酮,即染料木素。WB-F5是一株能产生高效抗菌活性物质的海洋放线菌,对WB-F5及其代谢产物进行研究,将有助于抗生素药物的开发研究。The anti-bacterial activity of marine actinomycete WB- F5 's broth was studied and its antibacterial substances were isolated and identified. The inhibitory activities of marine actinomycete WB- F5's broth against many kinds of bacterial were determined by agar punch method. The macro-reticular resin, the silica gel column chromatography and other methods were used to isolate the anti-bacterial substances. WB- F5 ' broth showed good inhibitory activities against Gram-positive bacteria including Staphylococcus aureus, MRSA Bacillus subtilis, and so on. According to the experiment results, the macro-reticular resin AB- 8 and the silica gel column chromatography were suitable for isolation. WBF5 - F was identified to be Genistein. Marine actinomycete WB - F5 can produce anti-bacterial materials which show high performance. The researche of WB- F5 and its metabolin will be helpful to the exploitation of antibiotics.
