摘 要:研究单环刺蜢纤溶酶(UFE—I)的溶栓作用及药物安全性。以家兔体外全血凝块溶解法和体内动-静脉旁路血栓模型法研究该酶的溶栓作用,通过分光光度法研究其溶血毒性,霍恩氏法研究其急性毒性。结果显示该酶体外溶栓活性强于20倍蚓激酶(EFE),具有较好的体内抗栓活性,无溶血毒性,对小鼠的半数致死剂量LD50为430mg/kg,95%的可信限为295-626mg/kg。该酶具有较强的溶栓作用和较高的安全性。To study the effect of fibrinolytic enzyme UFE- I from Urechis unicinctus on thrombolysis and its drug safety, the thrombolytic effect of UFE - I was observed in vitro with whole blood thrombus from rabbits and in vivo by the arteriovenous shunt model in rabbits, the hemolytic toxicity was investigated by spectrophotometry and the acute toxicity test was performed in mice by Horn's method. The results showed that UFE - I had apparent thrombolytic effects both in vitro and in vivo, and had no hemolytic toxicity. LD50 of UFE - I in mice was 430 mg/kg, and its 95% confidence interval was 295-626 mg/kg, and UFE- I possessed effective thrombolytic activity and high drug safety.
