在我国北方涉县首次检测到盖塔病毒

摘　　要：目的:确定从河北省涉县蚊体中分离到的病毒种类。方法:采集三带喙库蚊标本,液氮运输和保存,10只为一组研磨后接种C6/36细胞分离病毒。病毒株分别采用乙醚试验、免疫荧光试验和分子生物学试验进行毒株鉴定。结果:采集到蚊标本1290只,标本处理后接种细胞47批,病毒分离结果得到13株阳性分离物(毒株),经乙醚试验和免疫荧光试验初步鉴定HB0215-3和HB0234两株病毒为披膜病毒科甲病毒属成员,再经RT-PCR、核酸序列测定和分析,证实两株病毒核酸与国际基因库中的AY702913和NC-006558盖塔病毒同源性最高,被进一步鉴定为盖塔病毒。结论:该研究从河北省涉县三带喙库蚊中分离到了盖塔病毒,这是我国内陆地区北方首次分离到。Objective：In order to identify the kind of the viruses isolated from mosquitoes collected in She County of Hebei Province.Methods： Culex tritaeniorhynchus samples were collected and stored in liquid nitrogen.A group of 10 mosquitoes were ground and inoculated onto C6/36 cell.Positive isolates were identified by aether test,IFA and RT-PCR.Results： We had collected 1290 Culex tritaeniorhynchus.We had got 13 cytopathic effect positive isolates from 47 groups of mosquitoes.Two of 13 positive isolates（HB0215-3,HB0234） were alphavirus of togaviridea after aether test and IFA.After RT-PCR,nucleotide sequence detecting and analysing,because the nucleotide sequence of the two isolates have the highest homology to Getah virus AY702913 and NC-006558 of the international GenBank,the two isolates were identified as Getah Virus.Conclusion： Getah Viruses were first isolated from the culex tritaeniorhynchus samples in She County of Hebei Province,North China.
