摘  要:目的探讨胰岛素样生长因子1(hIGF-1)转染骨髓间充质干细胞(MSCs)对急性心肌梗死大鼠心功能的影响。方法雌性Wistar大鼠30只,建立急性大鼠模型随机分为3组:对照组。冠脉结扎后注射无血清培养液;MSCs组,冠脉结扎后注射MSCs;hIGF-1+MSCs组,冠脉结扎后注射转染hIGF-1的MSCs。细胞移植4周后利用心动超声检测各组大鼠心肌短轴缩短率(FS)及射血分数(EF);利用TUNEL法检测各组的凋亡指数。结果对照组、MSCs组和hIGF-1+MSCs组的FS分别为(0.27±0.03)、(0.36±0.07)和(0.46±0.09),EF分别为(0.47±0.05)、(0.63±0.10)和(0.76±0.13);与对照组相比,MSCs组和hIGF-1+MSCs组的FS和EF差异均有统计学意义(P〈0.05);与MSCs组相比,hIGF-1+MSCs组的FS和EF差异均有统计学意义(P〈0.05)。TUNEL法检测凋亡指数表明,与对照组相比,MSCs组和hIGF-1+MSCs组差异均有统计学意义(P〈0.05)。结论hIGF-1联合MSCs移植可以通过抗凋亡明显改善大鼠心功能。Objective To investigate the effect of bone marrow mesenchymal stem cells(MSCs) with hIGF - 1 transplantation on the cardiac function in rats with acute myocardial infarction. Methods Thirty Wistar rats were randomly divided three groups 10 rats in each group. The contrast group received DMEM/F12 after left coronary artery ligation to induce acute myocardial infarction, the MSCs group received myocardial injection of MSCs and ( hlGF - 1 ) MSCs group received myocardial injection of (hIGF - 1 ) MSCs after coronary artery ligation. After four weeks, cardiac function and heart tissue pathological changes were observed. Results left ventricular ejection fraction (EF) is ( 0. 47 ±0. 05 ) (contrast group), (0. 63 ±0. 10) ( MSCs group) and(0. 76±0. 13 ) ( hlGF - 1 ) MSCs. Left ventricular fractional shortening rate (FS) is ( 0. 27± 0. 03 ) ( contrast group), ( 0. 36 ± 0. 07 ) ( MSCs group) and (0. 46 ±0. 09) ( hIGF - 1 ) MSCs and the cardiac function of ( hIGF - 1 ) MSCs group was increased as compared with contrast and MSCs group ( P 〈 0. 05 ). apoptosis index show ( hIGF - 1 ) MSCs is better than MSCs in anti - apoptosis (P 〈 0. 05 ). Conclusion transplantation of MSCs with hIGF - 1 can improve the cardiac function in rat model of acute myocardial infarction by anti - apoptosis.
