摘  要:目的:用高效液相色谱(HPLC)法与超高效液相色谱(UPLC)法对比测定千里香药材中九里香酮的含量。方法:分别建立了HPLC和UPLC测定千里香药材中九里香酮含量的方法。HPLC法为AKZONOBEL Kromasil 100-5C18(4.6mm×250mm,5μm)色谱柱,以乙腈-水(30∶70)为流动相,流速1mL.min-1,检测波长337nm,柱温30℃;UPLC法为WatersAcquityBEHC18(2.1mm×50mm,1.7μm)色谱柱,以乙腈-水(23∶77)为流动相,流速0.5mL.min-1,检测波长337nm,柱温30℃。结果:2种方法所得含量测定结果一致。HPLC法和UPLC法加样回收率的RSD分别是1.8%和1.7%。结论:HPLC和UPLC法测定千里香中九里香酮的含量,方法简便,结果准确可靠。UPLC较HPLC法更加简便迅速,既达到了分析的高通量,得到更高的分析灵敏度,又大大提高了分析速度,减少了有机溶剂的消耗。UPLC法能够成功替代HPLC法,为千里香药材质量控制提供新的检测方法。Objective:To compare the methods—HPLC and ultra performance liquid chromatography(UPLC) for the determination of 6,8,3',4',--tetramethoxy flavone in Murraya paniculata (L.) Kack.Method:Respectively establish the HPLC and UPLC methods for the determination of 6,8,3',4',--Tetramethoxy flavone in Murraya paniculata (L.) jack.A AKZONOBEL Kromasil 100-5 C18column (4.6 mm×250 mm,5 μm)was adopted with methanol-water (30:70)as mobile phase at a flow rate of 1 mL·min-1;detective wavelength was set at 337 nm and column temperature was 30 ℃ in HPLC.A Waters Acquity BEH C18 column(2.1 mm×50 mm,1.7 μm)was adopted,with acetonitrile-water (23:77)as mobile phase at a flow rate of 0.5 mL·min-1;detective wavelength was set at 337 nm and column temperature was 30 ℃ in UPLC.Results:Both of the two methods for the determination had the same results.The RSD of the average recovery of HPLC and UPLC were 1.8% respectively,1.7%.Conclusion:HPLC and UPLC methods for the determination of 6,8,3',4',--tetramethoxy flavone in Murraya paniculata (L.) Jack are simple,the results are accurate and reliable.The major advantages of UPLC,over HPLC are the speed of analysis,the narrower peaks(giving increased signal-to-noise ratio),UPLC can not only increase separation speed and efficiency,but also reduce solvent consumption and analysis time.UPLC as a new detection method can replace HPLC for the quality control of Murraya paniculata (L.) Jack.
