摘　　要：目的探索ＣＤ４０反义ＲＮＡ对肿瘤传代细胞Ｋ５６２细胞株的影响。方法用本室构建的ＣＤ４０反义ＲＮＡ转导Ｋ５６２细胞。结果发现转导反义ＲＮＡ后Ｋ５６２细胞体积缩小，代谢变慢，光镜下见细胞核固缩或碎裂成块状；其ＤＮＡ电泳呈梯状断裂现象，用ＣＤ４０基因特异性引物和寡核苷酸探针进行ＲＴ－ＰＣＲ和Ｓｌｏｔｂｌｏｔ方法检测，发现Ｋ５６２细胞为ＣＤ４０基因阳性。结论Ｋ５６２细胞为ＣＤ４０基因阳性细胞。Objective To explore the effects of CD40 anti sense RNA on K562 cell line.Method The authors constructed CD40 anti sense RNA and transfected into K562 cell line using lipofectamine method.Result The results showed that K562 cells were become condense in nuclus and dark staining in Giemsa staining after transfecting with CD40 antisense RNA.The DNA of these transfected K562 cells showing ladder patttern in 1.5% agrose elecyropheresis.K562 cells were found that CD40 gene positive in RT PCR and slot blot assaying.Conclusion The results suggest that K562 cell is CD40 positive and affected by CD40 antisense RNA.
