摘 要:目的:探讨冬凌草乙素对U937细胞的生长抑制作用及其作用机制。方法:以不同浓度的冬凌草乙素作用于体外培养的U937细胞,MTT法检测细胞生长抑制率,应用hoechst33258荧光染色法观察细胞凋亡;采用RT-PCR对细胞凋亡前后β-catenin的表达水平进行检测。结果:冬凌草乙素可显著抑制细胞的生长及诱导细胞发生凋亡,呈现出明显的量-效与时-效关系。在冬凌草乙素抑制细胞增殖过程中β-catenin的表达水平显著下降。结论:冬凌草乙素能抑制U937细胞的生长并诱导细胞发生凋亡,降低β-catenin的表达水平可能是其重要作用机制之一。这为冬凌草乙素抗白血病的进一步研究提供了有力的实验依据。Objective To investigate the anti-proliferation effect of Ponicidin (PON)in leukemic U937 cells and its influence on β-catenin expression. Methods U937 cells in culture medium in vitro were treated by different concentrations of PON. The inhibitory ratio of the cells were measured by MTT assay, apoptosis was obsevered by hoechst 33258 fluorescence staining, and β-catenin expression was detected by RT-PCR. Results PON could inhibit the growth of U937 cells and cause apoptosis remarkably, the suppression was both in time- and dose-dependent manner, and β-catenin expression was downregulate significantly after PON treatment. Conclusion PON can inhibit the growth and induce apoptosis on U937 cells significantly. Downregulating of β- catenin expression of U937 cells may be one of its important mechanisms. The results provide strong evidence for further anti-leukemia investigation for PON.
