摘 要:目的:研究血清钙调神经磷酸酶(CaN)和正常T细胞表达和分泌的活性调节蛋白(RANTES)水平与兔动脉粥样硬化斑块易损性的关系。方法:30只雄性家兔随机分为:(1)空白对照组(Control组,10只),普通饲料喂养12周;(2)稳定斑块组(AS组,10只),高脂饮食(含胆固醇1%和猪油5%)喂养12周;(3)易损斑块组(VAP组,10只)高脂饮食喂养12周,于处死前24和48h分别给予两次药物触发。测定各组第0周、12周血脂,12周处死后检测兔血清CaN和RANTES水平,Masson染色法计算校正的斑块面积、易损指数及其与血清细胞因子的相关系数。结果:高脂喂养12周后AS组和VAP组的血脂、CaN水平较Control组明显升高(P<0.01)。VAP组RANTES水平(97.48±12.22)ng/mL明显高于AS组(25.01±4.45)ng/mL和Control组(24.88±5.22)ng/mL(均P<0.01)。与AS组比较,VAP组校正的斑块面积[(52.26±11.58)%vs(32.26±9.76)%]和易损指数(6.25±1.97vs3.24±1.12)差异均有显著性(P<0.01);AS组和VAP组的RANTES水平与两个易损指标均明显正相关(P<0.05);AS组CaN水平与两个易损指标均呈负相关(r=-0.62,-0.67;P<0.05)。结论:RANTES可能是易损斑块预测和干预的重要指标。Objective To explore the association of serum levels of calcineurin (CaN) and regulated upon activation normal T cell expressed and sec reted factor (RANTES) with the vulnerability of atherosclerotic plaques in rabbits. Methods 30 male rabbits were randomized to receive a standard diet for 12 weeks (control group, n = 10), a high-cholesterol diet (1% cholesterol plus 5% lard) (AS group, n = 10), or a high-cholesterol diet (VAP group, n = 10). All the VAP group rabbits were triggered twice by drugs 24 and 48 h before execution. Levels of blood lipids were detected at weeks 0 and 12 and serum levels of CaN and RANTES were measured at week 12. The adjusted plaque area and vulnerability index were calculated by Masson staining and its correlation coefficient with cytokines were measured. Results Levels of blood lipids and CaN level were increased significantly in AS group and VAP group than in control group (P 0.01); RANTES level was markedly higher in VAP group than in AS group and control group (97.48 ± 12.22) ng/mL vs (25.01 ± 4.45) ng/mL vs (24.88 ± 5.22) ng/mL (P〈 0.01); the adjusted plaque area and vulnerability index were obviously higher in VAP group than in AS group [(52.26 ± 11.58)% vs (32.26 ± 9.76)%] and (6.25 ± 1.97) vs (3.24 ± 1.12), P 〈0.01; RANTES level was positively related with the adjusted plaque area and vulnerability index in VAP group and AS group (P 〈0.05); CaN level was negatively related with the adjusted plaque area (r = -0.62, P 〈0.05) and vulnerability index (r = -0.67, P 0.05) in AS group. Conclusion RANTES may be an important indicator for predicting and preventing vulnerable atherosclerotic plaques.
