摘 要:目的:探讨胰岛素类似物对人脐带干细胞增殖活性的影响。方法:分别采用3种不同浓度(1、10、100nmol/L)的人胰岛素、门冬胰岛素和甘精胰岛素作用于体外培养的人脐带间充质干细胞,于培养的24、48和72h,采用MTT法检测细胞的增殖活性。结果:与对照组比,10、100nmol/L人胰岛素、门冬胰岛素和甘精胰岛素作用24h后脐带干细胞的增殖活性均明显增加(P<0.05);与对照组比,1、10、100nmol/L的人胰岛素、门冬胰岛素和甘精胰岛素作用48、72h脐带干细胞增殖活性均明显增加,且促细胞增殖效应各组均呈剂量依赖性,在高浓度组最明显;与人胰岛素组比较,门冬胰岛素组细胞增殖活性无明显增加(P>0.05),甘精胰岛素组在培养48h(P<0.05)和72h(P<0.01)细胞增殖活力显著强于人胰岛素组。结论:人胰岛素和胰岛素类似物均能促进脐带干细胞的增殖活性,门冬胰岛素促进干细胞增殖活性与速效胰岛素相当,甘精胰岛素促细胞增殖的能力明显强于人胰岛素,这为脂肪组织工程获得足够的种子细胞提供了一种新途径。Objective To Investigate the effect of insulin analogue on the proliferation of human umbilical cord stem cells. Methods Human umbilical cord mesenchymal stem cells were incubated with 1,10 and 100 nmol / L of regular human insulin, insulin aspart and insulin glargine, respectively. Cell proliferation was detected by MTT assay after 24-hour, 48-hour and 72-hour culture. Result Regular human insulin, insulin aspart and insulin glargine, at dose of 10 nmol / L and 100 nmol / L, could significantly increase cell proliferation compared with control group after 24-hour culture. Human insulin, insulin aspart and insulin glargine, at dose of 1, 10 and 100 nmol / L, could significantly increase cell proliferation compared with control group after 48-hour and 72-hour culture. Human insulin, insulin aspart and insulin glargine could promote cell proliferation in a dose- dependent manner. Insulin aspart showed no significant effect on cell proliferation compared with human insulin(P 0.05). Insulin glargine showed significant effect on cell proliferation compared with human insulin after 48- hour culture (P 0.05) and 72- hour culture (P 0.05), respectively. Conclusion Both regular human insulin and insulin analogues can enhance human umbilical cord stem cell proliferation. Insulin aspart and regular human insulin have similar effects on cell proliferation, Cell proliferation effect of insulin glargine was stronger than that of human insulin. This study may provide a new way to prepare sufficient seed cells for adipose tissue engineering.
