RT-PCR结合地高辛标记定量检测猴免疫缺陷病毒RNA

摘　　要：从感染猴免疫缺陷病毒（ＳＩＶ）的恒河猴血浆和培养细胞株中分别纯化病毒ＲＮＡ，经逆转录－聚合酶链反应（ＲＴ－ＰＣＲ）扩增ＳＩＶｇａｇ基因特异的ＤＮＡ片段。将ＲＴ－ＰＣＲ产物用地高辛标记，检测灵敏度比凝胶电泳和斑点杂交提高１００倍。用已知拷贝数的ＲＮＡ作为定量标准，可估计待测样本中ＳＩＶＲＮＡ的拷贝数。A simian immunodeficiency virus(SIV)gag gene_specific DNA fragment was amplified by reverse transcription-polymerase chain reaction(RT-PCR)of RNA purified from the serum of SIV-infected marcaque monkey and a cultured cell line. The detection sensitivity of RT-PCR products by digoxin labeling was 100 folds as that by gel electrophoresis and dot blotting. The copy number of SIV RNA to be measured could be estimated by using a standard RNA whose copy number was known.
