摘　　要：目的研究Ａｓ２Ｏ３在体外对人类Ｂ淋巴瘤细胞株ＭＢＣ－１细胞增殖的影响，并探讨其作用机制。方法采用细胞形态学、ＤＮＡ凝胶电泳、流式细胞术等多参数观察。结果（１）（０．５～２）ｍＭ／ＬＡｓ２Ｏ３对ＭＢＣ－１细胞有增殖抑制作用，并呈时间和剂量相关；（２）证实诱导细胞凋亡是Ａｓ２Ｏ３增殖抑制作用的主要机制之一；（３）Ａｓ２Ｏ３能够上调ＭＢＣ－１细胞ｐ５３的蛋白表达。结论Ａｓ２Ｏ３能有效地通过诱导人类Ｂ淋巴瘤细胞株ＭＢＣ－１细胞凋亡而起到增殖抑制作用，而ｐ５３基因可能参与了对该凋亡过程的调控。Objective In this present study,an investigation was made to determine the effect of Arsenic Trioxide(As 2O 3) on proliferation of the human B lymphoma cell line MBC 1. Methods Cell morphology,DNA electrophoresis, flow cytometry were used. Results The results showed that As 2O 3 could inhibit the proliferation of MBC 1 cells. With the morphologic observation, flow cytometry assay and DNA gel electrophoresis,we can confirm that this inhibition role was induced through apoptosis. And we also find out that As 2O 3 can upregulates p53 gene expression at protein level. Conclusion It is suggested that induction of cell apoptosis can be one of the mechanisms in the growth inhibition of As 2O 3 in MBC l cells. Upregulation of p53 expression appears to play an important role in the apoptosis of MBC 1 cells.
