摘 要:目的:研究中药胃肠舒对糖尿病(diabetes mellitus,DM)大鼠胃窦干细胞生长因子及其受体(stemcellfactor-kit,SCF-kit)和神经递质的影响,探讨胃肠舒促胃肠运动的机制。方法:标准SD大鼠随机分为:正常对照组、DM组、DM+胃肠舒Ⅰ组、DM+胃肠舒Ⅱ组。流式细胞仪检测各组大鼠胃窦组织中Cajal间质细胞(interstitial cells of Cajal,ICC)的数量,RT-PCR、Western blot分别测定SCF-kit mRNA及蛋白水平上的表达,免疫组化法、激光共聚焦显微镜分别检测神经递质及Ca2+的变化。结果:胃肠舒可增加DM大鼠胃窦ICC的数量,上调SCF-kit mRNA及蛋白表达,降低NO的表达,Ach、Ca2+表达增高。结论:胃肠舒可能通过改变SCF-kit和神经递质的表达,改善了ICC,从而促进胃肠运动。Objective:To study the mechanism of the Chinese medicine Weichangshu in the regulation of gastrointestinal motility propulsion by measuring the changes of SCF-kit and neurotransmitters in diabetic mellitus rats.Methods:Standard Sprague Dawley rats were randomly divided into 4 groups,including a control group,DM model group,DM+WeichangshuⅠ group,DM+WeichangshuⅡgroup.The amount of interstitial cells of Cajal was detected by flow cytometry.SCF-kit mRNA and protein expression were determined with RT-PCR and Western blot.Expression of neurotransmitters was observed via immunohistochemistry.Ca2+concentration was detected by laser scanning confocal microscope.Results:Weichangshu increased the amount of ICC.SCF-kit mRNA and protein were significantly up-regulated.Expression of NO decreased.Expression of Ach and Ca2+concentration increased.Conclusion:Weichangshu probably affected ICC through regulating SCF-kit and neurotransmitters,then promoted the gastrointestinal motility.
