不同介入时机针刺三阴交穴对痛经大鼠下丘脑、垂体β-EP含量和HSP70表达的影响研究

摘　　要：目的:探讨即刻针刺和逆针都可以减轻痛经模型大鼠疼痛的作用机制。方法:通过阴道涂片法,筛选出处于动情间期3月龄SD雌性大鼠,按照介入时机随机分为模型A组、盐水A组、针刺A组(即刻针刺组)和模型B组、盐水B组、针刺B组(逆针组),共6组,每组16只,观察不同介入时机(即刻、逆针)针刺三阴交,痛经大鼠下丘脑、垂体β-EP含量和HSP70表达的变化。结果:即刻针刺对痛经大鼠下丘脑、垂体β-EP的影响不显著;模型B组下丘脑β-EP含量与盐水B组相比有极显著性差异(P<0.01);逆针可以使下丘脑、垂体β-EP显著升高(P<0.05)。模型A组与盐水A组下丘脑HSP70表达有显著性差异(P<0.05)。结论:即刻针刺和逆针都有调整痛经模型大鼠中枢β-EP的合成与释放,使紊乱的下丘脑HSP70表达趋于正常的作用,但逆针的调整作用更显著,这可能是即刻针刺和逆针减缓痛经程度的中枢镇痛和应激机制之一。Objective：To investigate the mechanism of acupuncture and preventive acupuncture in alleviating the pain of dysmenorrhea model rats.Methods：96 Three month-aged SD female rats examined by vaginal smear were chosen and divided into six groups to observe the change ofβ-EP and HSP70 in hypothalamus and pituitary of dysmenorrhea rats with acupuncture and preventive acupuncture on Sanyinjiao point.Results：After modeling,hypothalamicβ-EP level,in dysmenorrheal group B was significantly increased（P0.01）.Preventive acupuncture can make the ccontent ofβ-EP in hypothalamus and pituitary increased significantly（P0.05）,but acupuncture was not.Model group A in hypothalamic HSP70 was significantly different to normal group A（P0.05）.Conclusion：Both acupuncture and preventive acupuncture can adjuste the synthesis and release of centralβ-EP in dysmenorrhea rats,make disordered hypothalamic HSP70 expression tends to normal,but preventive acupuncture against a significant role,which may be one of the central pain aand stress mechanisms of acupuncture and preventive acupuncture can slow down the degree of dysmenorrhea.
