摘  要:目的:建立一种快速测定星形胶质细胞上清中兴奋性氨基酸释放的高效液相-串联四级杆质谱方法。方法:使用高效液相-串联四极杆质谱的多反应监测(MRM)模式快速测定星形胶质细胞(AST)兴奋性氨基酸(EAAs)的释放量。结果:该方法检测细胞上清液中兴奋性氨基酸释放量能够在4分钟内完成,有很好的精密度(Glu1.0%,Asp1.6%),较好的稳定性(组内的变异系数Glu为1.34%,Asp为1.41%;组间变异系数Glu为2.32%,Asp为2.18%)和回收率(Glu97.1%;Asp95.8%)。结论:该方法定量检测兴奋性氨基酸快速、准确,可用于大批量样品的快速测定。Objective: To establish a method for determination of excitatory amino acids release of astrocytes activated with TNF-α by HPLC triple-quadrupole tandem mass spectrometer. Methods: Using HPLC-MS/MS to determinate the excitatory amino acids release of astrocytes by multi-reactions monitoring(MRM) technology. Results: Compared with the former methods(beyond 30min), the determination can be done in four minutes, the method had great precision (Glu 1.0%,Asp 1.6 % ), better stability (intragroup RSD Glu 1.34%, Asp 1.41%; interclass RSD Glu 2.32% ,Asp 2.18%), high average recoveries (Glu 97.1%;Asp 95.8%). Conclusions: The method had the advantages of rapidness, higher precision and higher sensitivity, was suitable for the determination of excitatory amino acids.
