摘  要:目的观察胡芦巴总皂苷(TFGs)对高糖高脂饮食诱导的肥胖大鼠血糖、血脂及胰岛素抵抗的影响。方法 Wistar大鼠30只分为正常组、模型组和治疗组,每组10只。模型组和治疗组给予高糖高脂饮食喂养16周后,治疗组给予TFGs(100mg/kg)灌胃,正常组和模型组给予等量生理盐水灌胃。治疗8周后,采用腹腔注射葡萄糖耐量试验(IPGTT)和胰岛素耐量试验(ITT)直接评价胰岛素抵抗水平,检测空腹血糖和胰岛素水平,计算胰岛素抵抗指数(HOMA-IR)间接评价胰岛素抵抗水平,同时测量体重、附睾周围脂肪重量,并检测血脂水平。结果 IPGTT、ITT和HOMA-IR结果显示模型组发生明显胰岛素抵抗,而经TFGs治疗后,治疗组胰岛素抵抗得到明显改善。同时,模型组较正常组大鼠体重、附睾周围脂肪重量、三酰甘油和总胆固醇水平明显升高,而治疗组较模型组明显降低。结论 TFGs能明显改善高糖高脂饮食诱导的肥胖大鼠胰岛素抵抗,同时能减轻体重,减少脂肪沉积,调节血脂代谢。Objective To investigate the effect of Trigonella foenum-graecum saponin (TFGs) on blood glucose and lipid metabolism and insulin resistance in obese rats induced by high-fat and high-sucrose diet. Methods Thirty Wistar rats were randomized to normal, model and treatment groups, with 10 in each. After 16 weeks' administration of high-fat and high-sucrose diet in the model group and treatment group, treatment group received TFGs (100 mg/kg) treatment by gavage, while normal and model groups were given saline by gavage with the same dose for 8 weeks. Intraperinoneal glucose tolerance test (IPGTT) and insulin tolerance test (ITT) were adopted to evaluate insulin resistance directly. Meanwhile, HOMA-IR was used for indirect evaluation of insulin resistance. Additionally, the body weight and epididymis fat weight as well as blood lipid level were measured. Result IPGTT, ITT and HOMA-IR showed obvious insulin resistance in model group, which was remarkably ameliorated after the treatment of TFGs in treatment group. Compared with normal group, body weight, epididymis fat weigh and blood levels of triglyceride and cholesterol were significantly higher in model group. The above indexes were notably decreased in treatment group. Conclusions TFGs can effectively ameliorate insulin resistance induced by high-fat and high-sucrose diet. It can also reduce body weight and fat accumulation, and modulate blood lipid metabolism.
