摘 要:目的研究双歧杆菌表面分子脂磷壁酸(LTA)对衰老小鼠胸腺细胞凋亡及Bcl-2、Bax蛋白表达的影响,探讨双歧杆菌抗衰老的分子机制。方法选择30只小鼠,将其随机分为衰老模型组、LTA治疗组和青年对照组,前两组用D-半乳糖建立衰老小鼠模型,LTA治疗组用双歧杆菌LTA治疗。用原位末端标记法检测胸腺细胞凋亡,用免疫组化法检测胸腺Bcl-2、Bax的表达,并计算Bax/Bcl-2的比值。结果与青年对照组相比,衰老模型组胸腺细胞凋亡指数显著升高,Bcl-2蛋白表达量显著降低,Bax蛋白表达量显著升高,Bcl-2/Bax比值显著降低,差异均有统计学意义(P<0.01)。LTA治疗组上述指标与青年对照组比较,差异无统计学意义(P>0.05)。结论双歧杆菌LTA能延缓胸腺细胞凋亡,使Bcl-2/Bax比值优化,具有抗衰老作用。Objective To study the effect of lipoteichoic acid(LTA)from bifidobacterium on apoptosis and Bcl-2、Bax protein expression in thymus gland of aging mice,probe into the molecular mechanism of bifidobacterium.Methods The mice was divided into aging model group、LTA group and young compare group.To build the mice model of aging by inject D-glactose in aging model group、LTA group and LTA group were treated with LTA,then obseve apoptosis with TdT-mediated dUTP nick end labeling and Bcl-2、Bax protein expression with immunohisto chemical method in thymus gland.Results Compared with young compare group,apoptotic index was increased(P0.01),protein expression of Bcl-2 was decreased(P0.01),expression of Bax was increased(P0.01)and Bcl-2 /Bax was decreased(P0.01)in aging model group.They weren,t all change in LTA group.Conclusion LTA can delay apoptosis in thymus gland and optimize retio of Bcl-2/Bax.LTA has function on antiaging.
