临床试剂rIL-2和实验试剂IL-2对γδT细胞体外扩增影响的研究

摘　　要：目的检测经临床试剂重组白细胞介素2(rIL-2)和实验试剂白细胞介素2(IL-2)体外辅助扩增后γδT细胞的细胞纯度及对肿瘤细胞的细胞毒性差异。方法应用梯度离心法分离10例正常人外周血单个核细胞,经IL-2和唑来膦酸(Zol)联合刺激γδT细胞进行扩增,采用流式细胞仪鉴定γδT细胞及其纯度,并检测其对肿瘤细胞的细胞毒性作用。结果在Zol存在的情况下,经实验用IL-2刺激的γδT细胞纯度最高达97.3%,经临床试剂rIL-2刺激的纯度为51.42%(P<0.05),且IL-2扩增后的γδT细胞对肿瘤细胞的细胞毒性作用较rIL-2明显。结论在Zol的存在前提下,IL-2能促使人外周γδT细胞迅速增生,达到实验所需要的细胞纯度,并对肿瘤细胞具有较强的细胞毒性作用,而rIL-2则不能。Objective To detect the differences of the cell purity and cytotoxicity to tumor cells of γδ T cell which were amplified by recombinant interleukin 2（rIL-2）used in clinical therapies and the laboratory reagents of interleukin 2（IL-2）in vitro.Methods Peripheral blood mononuclear cells（PBMCs）were co-stimulated by rIL-2/IL-2 and Zoledronic acid（Zol）which were centrifugated gradiently from 10 cases of normal person peripheral blood.The γδ T cell with its purity was detected by flow cytometry and the cytotoxicity to tumor cells by γδ T cell was also considered.Results In the presence of Zol,the purity of amplified γδ T cells stimulated by IL-2 can reach 97.3%,which may achieve 51.42% by rIL-2（P0.05）.Stronger cytotoxicity to tumor cells was detected by amplified γδ T cells by IL-2.Conclusion With the existence of Zol,IL-2 could promote human peripheral γδ T cells proliferating quickly.The purity of γδ T cells had strong cytotoxicity function to the tumour cell,and could meets the experimental needs in vitro,but rIL-2 couldn＇t.
