摘  要:评估新型骨组织工程支架聚消旋乳酸/聚乳酸-聚乙二醇-聚乳酸(PDLLA/PLA-PEG-PLA)共混物复合重组人骨形态发生蛋白-2(rhBMP-2)后修复兔下颌骨缺损的效果。20只成年新西兰兔双侧下颌骨下缘形成15 mm×6mm全层骨质缺损,复合rhBMP-2的实验材料植入一侧缺损,未复合rhBMP-2的实验材料作为阴性对照。结果显示实验侧成骨时间早,呈多中心成骨,至12周时缺损基本修复。对照侧未见多中心成骨现象,缺损未能完全修复。结论认为聚消旋PDLLA/PLA-PEG-PLA共混物在体内具有传导骨形成功能,复合rhBMP-2后有诱导-传导双重功能,是一种很有前途的骨组织支架材料。This research sought to asses the efficacity of a new type of tissue engineering bone developed by PDLLA/PLA-PEG-PLA and BMP as a kind of bone graft substitute in the rabbit model of mandibular defects;15mm×6mm bilateral mandibular periosteum bone defects were made surgically in 20 New Zealand adult rabbits.The porous scaffolds impregnated with rhBMP-2 were used for the purpose,and the scaffolds without rhBMP-2 were used as control.The methods adopted in this research were:macroscopy,histomorphologic exam,X-ray exam,SEM micrography,computer-aided analysis and graphics.The experimental group was shown to have an earlier inception of bone forming.New bone formation was seen along the border of the original mandibular bone and in the middle.At 12 weeks after surgery,the defects were almost filled with new bone.In the control group,the defects could not be repaired in its entirety,and there was no new bone in the middle.The porous scaffold is a promising carrier for BMP.This kind of bone graft substitute can serve as an osteoconductive and osteoinductive matrix.
