摘 要:目的探讨人脐静脉内皮细胞株(ECV304)缺氧再给氧(H/R)损伤的发生机制及三七总皂苷(tPNS)对该病理生理过程的影响。方法将体外培养的ECV304分为七组,正常组;模型组;tPNS高、中、低浓度组;维拉帕米组;吡咯烷二硫氨基甲酸酯(PDTC)组。对各组进行相应处理后,分别检测各组ECV304超氧化物歧化酶(SOD)、谷胱甘肽(GSH)、丙二醛(MDA)的含量、核转录因子-B(NF-κB)及细胞间黏附分子-1(ICAM-1)的表达。结果模型组,tPNS高,中、低浓度组、维拉帕米组及PDTC组细胞氧自由基清除功能明显低于正常组,NF-κB及ICAM-1表达明显高于正常组,其中tPNS高、中、低浓度组细胞氧自由基清除功能明显高于H/R组,NF-κB及ICAM-1表达率明显低于H/R组,且呈浓度依赖性改变,tPNS高浓度组细胞氧自由基清除功能明显高于维拉帕米组及PDTC组,NF-κB及ICAM-1表达率明显低于维拉帕米组及PDTC组同期。结论 H/R可激活ECV304,使细胞氧自由基清除功能明显降低,NF-κB及ICAM-1的表达明显升高;tPNS能有效改善细胞氧自由基清除功能,降低NF-κB、ICAM-1活性,减轻H/R损伤,且高浓度tPNS效果优于维拉帕米、PDTC。Objective To study the mechanisms of ECV304 with hypoxia/reoxygenation(H/R) injury and the effect of total panax notoginseng saponins(tPNS) on the pathophysiologic process.Methods The cultured ECV304 were divided into 7 groups:Control group,H/R group,tPNS group(high,middle and low concentrations),verapamil group,pyrrolidine dithiocarbamate(PDTC) group.The contents of superoxide dismutase(SOD),glutathione(GSH)and malondialdehyde(MDA)in ECV304 were measured.The nuclear factor kappa-B(NF-κB)and intercellular adhesion molecule-1(ICAM-1)expression of ECV304 were detected.Results The content of SOD and GSH in H/R group,tPNS group,verapamil group,and PDTC group were significantly lower than that in control group.The levels of MDA,NF-κB and ICAM-1 in H/R group,tPNS group,verapamil group,and PDTC group were significantly higher than that in control group.The content of SOD and GSH in tPNS group were significantly higher and the levels of MDA,NF-κB and ICAM-1 were significantly lower than that in H/R group.The higher concentration of tPNS was more effective.The GSH in high concentrations of tPNS group was significantly higher than that in verapamil group and PDTC group.The levels of MDA,NF-κB and ICAM-1was significantly lower than that in verapamil group and PDTC group.Conclusion The results suggested that H/R could activate ECV304,increase the expression of MDA,NF-κB and ICAM-1.The tPNS could inhibit the expression of MDA,NF-κB and ICAM-1 efficiently and alleviate H/R injury.The high concentration of tPNS was more efficiently than verapamil and PDTC.
