摘  要:通过大孔树脂D-101吸附法提取雀稗麦角菌发酵液中麦角总碱粗品。采用硅胶薄板层析确定粗品组分及洗脱条件,进行碱性硅胶柱低压层析,单因素实验确定洗脱溶剂的比例及洗脱次数,最终分离纯化得到麦角总碱,计算总回收率。结果表明:总生物碱回收率为77.6%。硅胶层析分离上样量与硅胶质量比为1∶20,去杂洗涤剂为氯仿:甲醇=5∶1(V/V)的混合液,依次洗涤5次。用氯仿-甲醇2∶1(V/V)的洗脱剂洗脱,得到麦角总碱;从雀稗麦角菌发酵液中提纯麦角碱法回收率及纯度较高,具有广阔的应用前景。A method was developed to separate and purify total ergot in the zymotic fluid of Claviceps paspali.Firstly,Claviceps paspali were absorbed in macroporous resin D101,next,they were eluted by absolute alcohol,and evaporated to dryness with rotary evaporation meter.Then,Chromatography with silica gel in H column lower pressure was preceded after determining the component and the conditions of elution with silica gel.Lastly,the proportion of eluent and times of elution were determined with a single factor test.The mass ratio of silica and sample is 20,after eluted with 5 BV methanols and 2 BV methanol five times.The recovery of the ergotic alkaloids can achieve 77.6%.It is a better method and can be used to separate and purify total ergot from Claviceps Paspali.
