摘  要:目的探讨低剂量辐射(LDR)预处理对小鼠异基因骨髓移植(allo-BMT)后急性移植物抗宿主病(aGVHD)和血清Th1/Th2相关细胞因子的影响。方法建立小鼠allo-BMT模型,根据LDR干预与否将30只BALB/c小鼠随机分为A组[6MV-X全身照射(TBI)]、B组(LDR+TBI)、C组(空白对照组)。移植后观察A、B两组的aGVHD症状和肝、小肠、皮肤的病理学变化;移植后第8天取血检测IFN-γ和IL-4的水平。结果与A组比较,B组的aGVHD症状积分高,aGVHD的发生程度较轻,且生存期长(P<0.05);与C组比较,A、B两组的IFN-γ水平均高、IL-4水平均低(P<0.05)、但以B组为著(P<0.05)。结论 LDR预处理可减轻aGVHD的严重程度,与LDR预处理可以减少肠道损伤,调节Th1/Th2相关细胞因子趋于平衡的作用有关。Objective To observe the effects of low dose radiation(LDR) on acute graft-versus-host disease(aGVHD) and Th1/Th2 related cytokines in rats after allogenic bone marrow transplantation(allo-BMT).Methods The rat model of aGVHD had been established by allo-BMT with donor derived T cells.30 rats were divided into three groups randomly according to whether or not using LDR: TBI group(group A),LDR pretreament before exposing to TBI(group B),blank control group(group C).The symptoms and pathological changes of liver and small intestine and skin of aGVHD were observed.The serum levels of both IFN-γ and IL-4 were determined by ELISA.Results The score of aGVHD in group A was lower than that in group B(P〈0.05).The levels of IFN-γ in both groups A and B were higher than those in group C(P〈0.05),the level of IFN-γ in group A was lower than that in group B(P〈0.05).The levels of IL-4 in both group A and B were all lower than those in nomal group(P〈0.05),the level of IL-4 in group A was higher than that in group B(P〈0.05).Conclusions LDR can alleviates the severity of aGVHD through reducing intestinal damage and balancing the Th1/Th2-related cytokines after allo-BMT.
