摘  要:目的:探讨积雪草酸对慢性髓系白血病细胞急变株K562增殖抑制,诱导凋亡的作用及其机制。方法:四甲基偶氮唑盐(MTT)法检测细胞增殖活性,Annexin-V/PI双标流式细胞术检测细胞的凋亡率,Hoechesst33258染色法检测细胞的形态学变化,RT-PCR检测积雪草酸对survivin和bcl-2mRNA表达的影响。结果:积雪草酸对K562细胞有明显的增殖抑制作用,尤其在30μmol.L-1以上增殖抑制效应更加显著。24,48h的IC50值分别为(36.2±4.7)μmol.L-1、(27.4±3.7)μmol.L-1。积雪草酸能诱导细胞发生凋亡,可见典型的凋亡形态,呈剂量依赖性。积雪草酸可使survivin、bcl-2mRNA的表达呈剂量依赖性降低。结论:积雪草酸能抑制K562细胞的增殖及诱导凋亡,呈剂量依赖性。积雪草酸的促凋亡机制可能与下调survivin和bcl-2的转录水平有关。OBJECTIVE To explore the effect of proliferation inhibition and investigate antitumor mechanism. METHODS The effect of asiatic acid on growth of cells was determined by MTT assay,the induction of apoptosis was studied by flow cytometry, the morphological changes of cells were evaluated by Hoechest33258 staining, the changes of surviving and bcl-2mRNA expression levels were analyzed by RT-PCR. RESULTS Asiatic acid apparently inhibited cell proliferation of K562 cells,and the effect of significant inhibition was started at the concentration of 30 μmol·L-1. The IC50 of cells were (36. 2± 4. 7)μmol·L-1 , (27. 4 + 3. 7)μmol·L-1 for 24 hours. Asiatic acid could induce apoptiosis in a dose-dependent manner. Cells treated for 24 h showed typical apoptosis features induced by asiatic acid. The expression of survivin, bcl-2mRNA were decreased in a dose-de pendent manner. CONCLUSION Asiatic acid is able to significantly inhibit the proliferation and induce apoptosis of K562 cells,in a dose〉 dependent manner. The pro apoptisis mechanism of asiatic acid is associated with up-regulation of survivin and bcl- 2 transcription level.
