摘  要:目的探讨2型糖尿病患者腺苷酸活化蛋白激酶(AMPK)_(α2)基因单核苷酸多态性(SNP)-rs2143754与糖脂代谢的关系。方法采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)和测序的方法,同时检测生化指标。结果糖尿病组(T2DM)与健康对照组(NC)的基因型构成比差异有统计学意义(P<0.05);T/T基因型糖尿病患者有较低的空腹胰岛素和餐后2小时胰岛素,较高的HOMA-IR(P<0.01),较高的TC、TG、LDL-C和较低的HDL-C(P<0.05);rs2143754基因多态性与糖尿病心血管并发症明显相关(P<0.01),T等位基因是心血管疾病的非独立危险因素。结论 rs2143754具有多态性频率分布,T/T基因型胰岛素抵抗加重,是糖尿病心血管病并发症非独立危险因素。Objective To evaluate the relationship between SNP-rs2143754 of AMPKα2 gene and glucolipid metabolism among Tianjin population with T2DM in China. Methods PCR-RFLP and sequencing were performed. Results There was significant difference in genotype constituent ratio between T2DM and NDM(P〈0.01). T/T genotype had lower fasting insulin and 2-h insulin, higher HOMA-IR (P〈0. 01), higher TC, TG, LDL-C, and lower HDL-C (P〈0. 05). There was significant association between the polymorphism of rs2143754 of AMPKα2 and diabetic cardiovascular complications (P〈0. 01). T allele was an independent risk factor for diabetic cardiovascular complication. Conclusions The polymorphism of rs2143754 exists among Tianjin population. The insulin resistance of T/T genotes aggravates, which is independent risk factor for diabetic cardiovascular complication.
