摘 要:目的探讨糖尿病鼠肾脏是否存在低氧状态及其与糖尿病肾病(DN)的关系。方法将雄性SD大鼠随机分为DN组和正常对照(NC)组,采用链脲佐菌素诱导DN大鼠模型,连续多时点检测尿白蛋白、血糖、血尿素和血肌酐,同时行肾脏常规病理检查,免疫组化检测肾脏缺氧诱导因子1α(HIF-1α)与血红素氧化酶1(HO-1)的表达变化,并分析其相关性。结果 HIF-1α和HO-1在NC组肾小管仅轻度表达;而在DN组表达明显增强(P<0.01),主要集中在皮质区和皮髓质交界区的肾小管,峰值在8~12周。HIF-1α和HO-1表达变化与尿白蛋白、肾脏肥大指数和肾小球体积呈正相关。结论糖尿病肾脏缺氧状态参与了DN的发生发展。Objective To investigate whether hypoxic conditions exist and its relations to pathologic changes in the kidney of streptozotocin-induced diabetic rats. Methods Sprague-Dawley male rats were divided into diabetic nephropathy (DN) group and control group. Urinary albumin, blood sugar, serum urea and creatinine were monitored at 2,4,8, 12 and 16 weeks. Renal pathologic changes and levels of hypoxia inducible factor-1α(HIF-1α) and hemooxygenase-1 (HO-1) were observed at the same time points. Results HIF-1α and HO-1 in kidney were outstandingly expressed in renal tubules of the cortex or the close area between cortex and medulla, and their staining degree were significantly stronger in DN group than in the control group (P〈0. 01), being maximal at 8 and 12 weeks. In DN group, a strong correlation between HIF-1α and HO-1 levels was found(r=0. 838, P〈0. 01), and their levels were correlated with urinary albumin,nephrohypertrophy index and glomerular volume. Conclusions Hypoxia in the diabetic kidney involves in development of diabetic nephropathy.
