脂联素基因表达与妊娠糖尿病关系

摘　　要：目的检测脂联素mRNA在妊娠糖尿病患者脂肪及胎盘组织中的表达及在妊娠糖尿病发病机制中的作用。方法用半定量逆转录聚合酶链反应(RT-PCR)技术检测60名正常产妇及60例妊娠糖尿病(GDM)患者剖宫产后脂肪及胎盘中脂联素基因在转录水平的表达。结果 GDM患者脂肪组织中脂联素mRNA表达量为(1.05±0.12),正常妊娠者为(1.28±0.14),GDM患者低于正常妊娠组,差异有统计学意义(P<0.01);脂肪组织中脂联素mRNA表达量与GDM胰岛素抵抗指标相关(P<0.01);GDM患者与正常妊娠患者胎盘组织中脂联素mRNA表达差异无统计学意义(P>0.05)。结论脂肪组织脂联素表达的减少可能是GDM患者胰岛素抵抗的重要原因;胎盘脂联素的表达与GDM及其胰岛素抵抗关系不明显。Objective To investigate the expression of adiponectin mRNA in adipose and placenta tissue among normal pregnant women and gestational diabetes mellitus（GDM） patients in order to evaluate the role of adiponectin in GDM.Methods Reverse trascription polymerase chain reaction（RT-PCR） was used to detect adiponectin expression level in adipose and placenta of 60 GDM cases and 60 normal controls after caesarean section delivery.Results The expression of adiponectin was significantly decreased in the adipose tissue of GDM cases campared to that of the normal controls（1.05±0.12 vs1.28 ±0.14,P〈0.01） Conclusion The results prove that the alteration of adiponectin mRNA expression in adipose play an important role in the pathogenesis of GDM.
