摘 要:目的:运用"桑苏二陈汤加味"治疗哮喘发作期Wistar大鼠模型,观察治疗前后大鼠一般生活状态、细胞因子等指标的变化,探讨桑苏二陈汤加味对哮喘发作期的疗效及作用机制,为临床应用桑苏二陈汤加味治疗哮喘提供实验依据。方法:雄性Wistar大鼠90只按随机数字表分为6组:空白组,桑苏二陈汤加味高剂量组、中剂量组、低剂量组,定喘汤组,模型组,每组15只。大鼠哮喘模型参照吕国平[1]等介绍的方法复制。观察治疗前后大鼠一般生活状态变化;测定大鼠血清中的ET-1、IL-1、IL-6、TNF-α、NO含量。结果:与空白组比较,模型组大鼠ET-1?IL-1?IL-6?TNF-α、NO含量显著升高,提示哮喘炎症存在。与模型组比较,治疗组炎症有减轻,ET-1、IL-1、IL-6、TNF-α、NO含量下降。与定喘汤组比较,桑苏二陈汤加味高、中剂量组ET-1、IL-1、IL-6、TNF-α、NO含量升高(P<0.01),桑苏二陈汤加味低剂量组无统计学意义。桑苏二陈汤加味高中低剂量组比较,低剂量组较高、中剂量ET-1、IL-1、IL-6、TNF-α、NO含量降低(P<0.05,P<0.01)。结论:桑苏二陈汤加味低剂量组能显著降低哮喘大鼠血清中TNF-α、ET-1、IL-1、IL-6、NO含量,有效控制气道炎症,从而改善气道重塑。Objective:To provide the experimental basis for doctors to make decision in choosing Modified Sangsu Erchen Decoction (MSED) treating Allergic asthma. Methods:90 adult male Wistar rats were randomly divided into 6 groups: high dose of MSED group, middle dose of MSED group, low dose of MSED group, Dingchuan decoction group, blank group, and model group. All groups of rats except blank group were modeled by the method reported by Lv Guoping. The change of the rats were observed and the level of ET-1、IL-1、IL-6、TNF-α、NO in serum were tested. Results:Compared with blank group, the level of ET-1、IL-1、IL-6、TNF-α、NO in serum of model group increased, implied inflammation exists in asthma model rats. Compared with model group, the level of ET-1、IL-1、IL-6、TNF-α、NO in serum of the therapy group decreased. Compared with Dingchuan decoction group, the level of ET-1、IL-1、IL-6、TNF-α、NO of the high dose and middle dose of MSED group increased(P0.01). Compared with high dose and middle dose group, the level of ET-1、IL-1、IL-6、TNF-α、NO of the low dose of MSED group decreased (P0.01). Conclusion:Low dose of MSED can decrease the level of ET-1、IL-1、IL-6、TNF-α、NO, control the inflammation of air way and improve the remodeling of airway.
