摘 要:本文观察了反义c-mybODN对黄体细胞hCG诱导的P和E。产生的影响,并探讨了与二了酰cAMP和verapamil的关系。结果发现,反义0.mybODN能呈剂量相关方式抑制黄体细胞hCG诱导的P和民的产生,同时使c.myb蛋白染色阳性的黄体细胞百分数下降:而无义tatODN没有相应的作用。当104mollL二丁酰cAMP与反义c.mybODN共同作用时能明显逆转反义C.mybODN对产生和c.myb表达的抑制作用。而10‘mollLverapmil与反义0.mybODN共同作用时,对反义0.mybODN对产生和0.myb表达的抑制作用具有协同效应。表明,0.myb与黄体细胞hCG诱导的P和产生密切相关;cAMP和Ca2”能通过调节c。The effects of c-myb antisense oligodeoxynucleotides (antisense c-myb ODN) on the hCG-induced progesterone and estradiol production of isolated rat luteal cells and their effect of cAMP andCa2+ were investigated. Antisense c-myb ODN could inhibit the hCG-induced progesterone andestradiol production in a dose-dependent manner, the percentages of luteal cells stained for c-mybprotein were also significantly decreased; while nonsense tat ODN had no similar effective.Futherexperiments showed that dbcAMP 10`-4mol/L could increase the percentage of luteal cells stained for c-myb protein, and increase the hCG-induced P and E, production. When dbcAMP was given incombination with antisense c-myb ODN, the inhibitory effect of antisense c-nlyb ODN upon hCG-induced P and E, production was reversed,the percentage of c-myb protein,positive luteal cells werealso increased.However, verapamil (l0-6 mol/L) had opposite effects on c-myb expression and hCG-induced P and E, production as compared with dbcAMP.The results implied that cAMP and Ca2+_might be involved in regulating the expression of c-myb protooncogene,in luteal cells and then regulateluteal cell function.
