摘 要:分离大鼠脾淋巴细胞培养24小时后,经10~1000μm芥子气染毒30min继续培养3d,以单细胞凝胶电泳法检测DNA损伤。结果显示中毒后淋巴细胞DNA迁移率和受损细胞百分率呈时间和剂量依赖关系;中毒4h,高、中剂量与对照组相比呈显著性差异(P<0.01)。提示单细胞凝胶电泳法检测DNA损伤用作评价芥子气细胞毒性指标的可能性。Isolated spleen lymphocytes of rat having been exposed to 10~1000μm mustard gas at 37℃ 30 minutes,DNA damage in lymphocytes at 0,4,24 and 72h postexposure were measured with SCG assay.The results not only showed that time dependent relationship and dose dependent relationship between mustard gas and DNA migration and the percentage of DNA damaged lymphocytes,but also a highly significant increase in DNA damage at 10 -3 M,10 -4 M after 4h postexposure compared with the control group.It's suggested that the method may be used as and indicator of cytotoxicity resulted from mustard gas,and also it may play a certain role in early diagnosis in mustard gas intoxication.
