消癥通络中药对糖尿病肾病大鼠胃排空的影响

摘　　要：目的观察消癥通络中药对糖尿病肾病大鼠胃排空的干预作用并探讨其可能机制。方法选择雄性SD大鼠50只,随机分为空白组、模型组、中药低剂量组、中药中剂量组、中药高剂量组,用左肾切除术加腹腔注射链脲佐菌素制作糖尿病肾病模型,中药低、中、高剂量组分别灌服消癥通络中药5.29g/(kg.d)、10.58g/(kg.d)、21.16g/(kg.d),空白组、模型组大鼠灌服相当剂量的生理盐水,连续6周。实验结束测定胃排空率、血清瘦素、胰高血糖素水平。结果与空白组比较,模型组大鼠胃排空率明显下降(P<0.05),血清瘦素、胰高血糖素水平明显升高;与模型组比较,消癥通络中药能明显改善糖尿病大鼠胃排空,明显降低大鼠血清瘦素、胰高血糖素水平(P<0.05);血清瘦素、胰高血糖素水平与胃排空率呈明显负相关。结论消癥通络中药能明显改善糖尿病肾病大鼠胃排空功能,其机制可能与降低大鼠血清瘦素、胰高血糖素水平有关。Objective To observe the effect and possible mechanism of the Chinese medicines resolving masses and dredging meridians on gastric emptying of rats with diabetic nephropathy. Methods Eighty SD male rats were randomly divided into 5 groups： control group, model group, low dose treated group, middle dose treated group, and high dose treated group. Models of diabetes were induced by removing the left kidney and injecting streptozotocin （STZ） solution into abdominal cavity. The rats in the treated groups were given Chinese medicines 5.29g/（kg·d）, 10.58g/（kg·d）, and 21.16g/（kg·d） respectively by intragastric administration, while the rats in the control and model groups were administered the same dose normal saline for 6 weeks. Gastric emptying rate, plas- ma leptin, and glucagon were detected at the end of the experiment. Results Compared with the control group, the gastric emptying rate in the model group was significantly lowered （P〈0. 05）, the level of leptin and glueagon was significantly increased. Compared with the model group, the Chinese medicines could significantly improve the gastric emptying rate and decrease the level of leptin and glucagon （P〈0.05）. There was a clear negative correlation among the level of leptin, glucagon and the gastric emptying rate. Conclusion The Chinese medicines could significantly improve the gastric emptying rate of the rats with diabetic nephropathy. Its possible mechanism might be related to decrease of the plasma leptin and glucagon.
