摘  要:目的探讨过氧化氢(H2O2)摄入后小鼠睾丸生精细胞环氧化酶-2(COX-2)和Caspase-3表达的影响。方法健康雄性昆明种小鼠30只,随机分为对照组10只和实验组20只。对照组小鼠喂食自来水,实验组小鼠以0.3 g.L-1H2O2(0.3 g.L-1剂量组)和3.0 g.L-1H2O2(3.0 g.L-1剂量组)溶液喂食。30 d时处死各组小鼠,取出其睾丸,免疫组织化学链霉亲和素-生物素酶复合物染色法检测其生精细胞COX-2和Caspase-3表达的变化。结果与对照组比较,0.3 g.L-1剂量组和3.0 g.L-1剂量组COX-2、Caspase-3表达均显著升高,差异有统计学意义(Pa<0.05,0.01),与0.3 g.L-1剂量组比较,3.0 g.L-1剂量组COX-2无显著升高,Caspase-3显著升高(P<0.01)。结论 HO经口摄入可造成小鼠睾丸生精细胞的损伤。Objective To explore the effect of hydrogen peroxide(H2O2) on the expression of cyclooxygenase 2(COX-2) and Caspase-3 in mouse testicular spermatogenic cell.Methods Thirty healthy male mice of Kunming Genus were divided into 2 groups randomy: control group(n=10),experimental group(0.3 g·L^-1 H2O2 group,n=10;and 3.0 g·L^-1H2O2 groups,n=10).Control group ingested tap water,and mice in experimental groups ingested 0.3 g·L^-1or 3.0 g·L^-1H2O2 solution.On the 30th day,testis were quickly taken out,fixed in the Zamboni fixation solution,conventionally produced frozen section and stained,the expression of COX-2 and Caspase-3 were detected.Results Compared with the control group,the expression of COX-2 and Caspase-3 in 0.3 g·L^-1group and 3.0 g·L^-1group were significantly increased(Pa〈0.05,0.01).The expression of COX-2 in 3 g·L^-1group was samilar to that in 0.3 g·L^-1group,and the expression of Caspase-3 in 3.0 g·L^-1H2O2 group was significantly increased compared with that in 0.3 g·L^-1group(P〈0.01).Conclusion Harmful of mouse testicular spermatogenic cell can be caused by feeding H2O2.
