HPLC-ELSD测定枸骨叶中的冬青苷Ⅱ的含量

摘　　要：目的：建立枸骨叶中的冬青苷Ⅱ的HPLC-ELSD含量测定方法。方法：采用Waters Syemmetry ShieldRP18色谱柱（4.6 mm×250 mm,5μm）,以甲醇-水为流动相梯度洗脱,流速1.0 mL.min-1,柱温35℃,蒸发光检测器检测。结果：冬青苷Ⅱ在1.00~40.0μg线性关系良好,R2=0.999 7。方法回收率101.3%,RSD 2.0%（n=6）。结论：该方法准确可靠,且重复性好,可用于测定枸骨叶中的冬青苷Ⅱ。Objective： To develop a HPLC-ELSD method for determination of ilexsideⅡin leaves of Ilex cornuta.Method： The separation was performed on a Waters Syemmetry Shield RP18 column（4.6 mm×250 mm,5 μm） and methanol-water was used as mobile phase in gradient elution.The flow was 1.0 mL·min-1,and the temperature was set at 35 ℃.Result： The calibration curve showed good linear ity in the test range（R2=0.999 7）.And the mean recovery was 101.3%,RSD was 2.0%（n=6）.Conclusion： The developed HPLC-ELSD method was accurate and reproducible,and can be used for the determination of ilexside Ⅱ in leaves of Ilex cornuta.
