摘 要:目的研究大豆异黄酮对前列腺增生大鼠模型的抑制作用及生长因子调控机制。方法采用sc睾酮法诱导大鼠前列腺增生。大鼠随机分为对照、模型组、大豆异黄酮(60、120、240 mg/kg)组;ig给药4周后处死大鼠,测定大鼠前列腺湿质量、前列腺指数,酶联免疫法测定血清胰岛素样生长因-子1(IGF-1)、表皮生长因子(EGF)、血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)、转化生长因子-β(TGF-β)水平,免疫组化法检测表皮生长因子受体(EGF-R)表达的变化情况。结果模型组大鼠前列腺湿质量明显增加,前列腺指数明显增加。与模型组相比,大豆异黄酮各组前列腺湿质量和前列腺指数明显下降,血清IGF-1、EGF、VEGF和bFGF水平均显著降低,血清TGF-β水平显著提高,前列腺上皮组织EGF-R表达显著下降,以120 mg/kg大豆异黄酮组效果最为明显。结论大豆异黄酮具有较好的抑制前列腺增生的作用,可能与其降低IGF-1、EGF、VEGF和bFGF水平,提高TGF-β水平,下调EGF-R表达有关。Objective To investigate the inhibitive effect of soybean isoflavone(SI) on benign prostatic hyperplasia(BPH) in rats and to study its regulation mechanism of growth factors.Methods Rats were induced to prostate hyperplasia by sc injection of testosterone and were randomly divided into normal control group,BPH model group,and three SI groups(60,120,and 240 mg/kg).Treated by ig administration four weeks later,the prostate wet weight and prostatic index(PI) were measured.The levels of IGF-1,EGF,VEGF,bFGF,and TGFβ in serum were determined by ELISA,and the change of EGF-R was detected by immunohistochemistry.Results The prostate wet weight and PI increased in BPH model group;The prostate wet weight and PI decreased in all SI dose groups,and the levels of IGF-1,EGF,VEGF,and bFGF in serum and the expression of EGF-R in prostatic epithelial tissue also decreased,compared to the model group.However,the level of TGF-β increased significantly.The effect of 120 mg/kg SI group was the most potent.Conclusion SI has good inhibition on prostate hyperplasia and its mechanism may be related to reducing the levels of IGF-1,EGF,VEGF,and bFGF,raising the level of TGF-β,and down-regulating the expression of EGF-R.
