摘 要:目的:观察巴戟天寡糖对大鼠心肌缺血再灌注损伤的影响。方法:将96只Wistar大鼠随机均分为假手术组、缺血再灌注模型组、阳性药物地奥心血康对照组和巴戟天寡糖0.7、1.4及2.8g/(kg.d)给药组,每日给予相应剂量的巴戟天寡糖及地奥心血康药液灌胃,假手术组、缺血再灌注模型组给予相应体积的蒸馏水灌胃,共7d。末次给药1h后,各组动物均接受开胸,除假手术组不结扎外,其他各组均结扎左冠状动脉前降支30min,再灌注90min。观察6组大鼠的心律失常评分、心肌损伤及梗死面积、心肌组织中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)和丙二醛(MDA)等的变化。结果:6组大鼠缺血期和再灌注期心律失常评分、心肌损伤面积和梗死面积及心肌组织中SOD、CAT、GSH-Px和MDA的含量差异有统计学意义(F=265.017、284.667、2589.641、1885.223、64.061、403.713、36.586和45.934,P均<0.001);与模型组相比,阳性药物对照组、巴戟天寡糖1.4、2.8g/(kg.d)给药组心律失常评分降低,心肌损伤及梗死面积缩小,心肌组织中SOD、CAT和GSH-Px酶活性升高,MDA含量降低(P<0.05)。结论:巴戟天寡糖有抗心肌缺血再灌注损伤作用,其机制可能与提高心肌抗氧化酶活性及减少脂质过氧化反应有关。Aim:To investigate the effects of the ethanolic extracts of morinda officinalis oligose (MOO) on myocardial ischemia reperfusion injury in rats.Methods:A total of 96 Wistar rats were randomly allocated into 6 groups (16 rats per group):sham operation(Sham) group,myocardial ischemia reperfusion(I/R) model group,positive control drug (DK) group,and three different doses of MOO [0.7,1.4,2.8 g/(kg·d)] groups.The rats in DK and three MOO groups were intragastricly infused DK suspension 0.7 g/(kg·d) together with MOO 0.7,1.4 or 2.8 g/(kg·d) accordingly for 7 d,while same volume of distilled water was infused intragastricly in Sham and I/R model groups.1 h after last intragastric infusion,the rats were subjected to open chest.Except for the Sham group,the left anterior desending coronary arteries (LAD) were firstly ligated for 30 mins and then unclamped for 90 mins in the other groups.After the procedure,scores of ventricular arrhymia,ischemic zone size,infarct size of myocardium,superoxide dismutase (SOD),catalase (CAT),glutathione peroxidase(GSH-PX),malonialdehyde(MDA) were measured in each rat.Results:The scores of ventricular arrhymia in the ischemia period and the reperfusion period,the ischemic zone size and the infarct size of myocardium and levels of myocardium SOD,CAT,GSH-Px and MDA among the groups were different,and there were obvious differences (F=265.017,284.667,2 589.641,1 885.223,64.061,403.713,36.586 and 45.934,P0.001).Compared with the I/R group,the scores of ventricular arrhymia and the infarct size of myocardium were significantly reduced,the activities of SOD,CAT,GSH-Px significantly increased,while MDA content significantly reduced in myocardium of DK group,MOO 1.4 g/(kg·d) group and MOO 2.8 g/(kg·d) group(P0.05).Conclusion:MOO could protect myocardium against ischemia reperfusion injury.This may be due to inhibition of free radical and subsequent lipid peroxidation.
