摘  要:目的:探讨人绒毛膜促性腺激素(HCG)对3T3-L1脂肪细胞过氧化物酶增殖体激活受体γ(PPARγ)及脂联素mRNA表达的影响。方法:分别用50、500和5000U/LHCG干预未分化脂肪细胞2、4和6d,未用HCG干预为空白对照组,PT-PCR测定PPARγmRNA的表达;用诱导分化液(1μmol/L地塞米松+0.5mmol/L3-异丁基-1-甲基黄嘌呤+10ng/L胰岛素)诱导3T3-L1脂肪细胞分化为成熟脂肪细胞,油红O染色鉴定后用5000U/LHCG干预成熟脂肪细胞6、12和24h,未用HCG干预为空白对照组,用RT-PCR测定PPARγ及脂联素mRAN的表达情况。结果:不同剂量HCG干预未分化3T3-L1脂肪细胞,与空白对照组相比,不同剂量干预组未分化脂肪细胞PPARγmRNA的表达量均增加,差异有统计学意义(F=19.823、21.352和23.519,P均<0.001);不同剂量HCG干预组组间比较,5000U/L剂量干预组PPARγ的表达量较50和500U/L剂量组增加,差异均有统计学意义(P均<0.05);50和500U/L剂量组比较,PPARγmRNA的表达量差异无统计学意义(P>0.05)。5000U/LHCG干预已分化3T3-L1脂肪细胞后,空白对照组、干预6、12和24h组相比,脂肪细胞PPARγmRNA的表达差异有统计学意义(F=26.907,P<0.001),脂联素mRNA的表达差异无统计学意义(F=0.066,P=0.976)。结论:HCG可能促进3T3-L1脂肪细胞的分化,但对脂联素的分泌无影响。Aim:To investigate effects of human chorionic gonadotropin(HCG) on adipocyte peroxisome proliferator-activated receptor γ(PPARγ) and adiponectin mRNA expression in 3T3-L1 adipocytes.Methods:The preadipocyte and the differentiated adipocyte were intervened by the high glucose Dulbecco's modified eagle media (DMEM) which contains the different concentrations of HCG.The reverse transcriptase PCR technique were used to measured the different expression of the PPARγ and adiponectin mRNA.Three different concentration of HCG (50,500,and 5 000 U/L) were applied to intervented the differentiating preadipocyte for 2,4,and 6 days.The blank control group was not intervented by HCG,and then the expression of the PPARγ mRNA was detected with RT-PCR.The concentration of 1 μmol/L dexamethasone and 0.5 mmol/L IBMX and 10 ng/L insulin was used to induce the 3T3-L1 preadipocyte differentiation into the mature fat cell,and the cell were stain by oil red O.Similarly the differentiated adipocyte for 6,12,and 24 hours under 5 000 U/L HCG medium were observed,the blank control group was not intervented by HCG,then the expression of the PPARγ and adiponectin mRNA was detected by RT-PCR.Results:The expression of the PPARγ mRNA in the undifferentiated 3T3-L1 preadipocyte stimulated by different dosage of HCG.There were significant differences among four groups at three time points on PPARγ mRNA expression (F=19.823,21.352,23.519,P0.001).Compared with the group of 50 or 500 U/L,the expression of PPARγ mRNA in the group of 500 U/L HCG increased significantly (P0.05).Compared with the group of 50 U/L HCG,the expression of PPARγ mRNA in the group of 500 U/L HCG had no significant difference (P0.05).The expression of the PPARγ mRNA in the differentiated adipocyte stimulated by 5 000 U/L HCG for 6,12,and 24 h had significant difference (F=26.907,P0.001),and the expression of adiponectin mRNA had no difference (F=0.066,P=0.976).Conclusion:The differentiation of 3T3-L1 adipocyte could be sti
