摘　　要：目的探讨Delta1基因转染DCs延长异基因大鼠心脏移植物存活的作用及其相关机制。方法供体SD大鼠心脏移植给受体Winstar大鼠后,经尾静脉注入1×106Delta1基因转染DCs。每天触摸受体大鼠腹壁,记录心脏移植物存活时间。受体大鼠尾静脉取血,采用酶联免疫吸附法(ELISA)测定Delta1蛋白的体内表达;通过混合淋巴细胞反应(MLR)及辅助性T细胞Th1/Th2型细胞因子检测,初步探讨耐受机制。结果经Delta1基因转染DCs处理的受体大鼠,异基因心脏移植物平均存活时间达(11.7±2.5)d,而对照的未处理组和DCs处理组平均存活时间分别为(6.7±1.7)d和(7.3±2.6)d,差异具有显著意义(P<0.05)。心脏移植物长期存活的受体大鼠MLR活性特异性降低,Th1/Th2型细胞因子向Th2方向偏移。结论 Delta1基因转染DCs使异基因大鼠心脏移植物长期存活。该效应为供体特异性,可能与Th1/Th2型细胞因子偏离有关。This p ape r is aimed to investigate the potential of Delta1 gene transfected dendritic cells （DCs） in inducing transplantation tolerance and its related mechanisms by using rat cardiac graft model. The heart allograft of SD rat was placed in the abdomen of Winstar rat,and Delta1 gene transfected DCs were infused at a dose of 1 ×106 via pigtail vein immediately after the operation. The survival of cardiac allografts was monitored by daily palpation and the total cessation of beating was defined as rejection and was confirmed by histology. The survival of allografts were prolonged significantly in recipients receiving Delta1 gene transfected DCs with a mean survival time of 11.7±2.5 d （n = 10）,comparing with those of the untreated recipients （6.7±1.7 d,n =10） and DCs treated recipients （7.3±2.6 d,n = 10） （P 0. 05）. And ELISA detected a prolonged expression of serum Delta1 in Delta1 gene transfected DCs treated rats. Furthermore,mixed lymphocyte reaction （MLR） displayed donor-specific hyporesponsiveness of splenocytes in recipients with long-term surviving cardiac allograft,while Th1/Th2 polarization analysis showed a dominance of Th2 type cytokines. We concluded that Delta1 gene transfected DCs prolong therapeutic expression of Delta1 in recipient rats,leading to long -term survival of cardiac allografts. And the induced tolerance is donor-specific,and may correlated with the deviation of Th1/Th2 type cytokines.
