摘  要:在室温自然光线下得到了干燥、水合及缓冲液内紫膜菌紫质的(?)(?)叶变换红外光谱及其二级导数谱提高了谱线的表与分辨,将原谱的重叠组分有效地分离开.讨论了原谱与二级导数的关系.对经分辨的各组分谱线作了识别.结果表明,紫膜菌紫质内除α—螺旋结构外,还存在相当量的β-折叠和转折结构,在各种水合条件下,各特征谱线无明显的频率位移.The Fourier transform infrared spectra and their second derivative spectra of bacteriorhodopsin have been recorded at room temperature and light in purple membranes which were P2O5-and air-dried, rehydrated over saturated K2SO4 solution, and in buffer. The second derivative procedures significantly enhace the resolution of infrared spectra of the protein in both the solid and solution states, seperating the overlapping components of the original spectra efficiently. The relationship of the original spectra to their second derivatives is discussed The resolved lines are assigned to the components of secondary structures of the protein. A large amount of transmembrane α-helices with some proportions of β-strands and turns. No detected sfifts of frequencies of the characteristic absorption bands have been observed at different conditions studied.
