摘 要:目的分离纯化海藻硫酸多糖,观察其对小鼠S180肿瘤的抑制效果,并从免疫调节和抗氧化的角度探讨其抗肿瘤作用机制。方法应用SephacrylS200凝胶柱检验所得海藻硫酸多糖组分均一性,以气相色谱技术研究其单糖组成;建立小鼠荷S180肿瘤模型,腹腔注射给药,观察抑瘤率、脾指数和胸腺指数,检测NK细胞杀伤活性、T淋巴细胞增殖以及肝脏和血清抗氧化活性。结果海藻硫酸多糖经SephacrylS200洗脱呈单一峰,说明组分比较均一,单糖组成以岩藻糖和半乳糖为主;低、中、高海藻硫酸多糖剂量组抑瘤率分别为4.89%、26.63%和29.35%,初步研究显示海藻硫酸多糖的抑瘤机制主要是免疫调节和抗氧化作用。结论所得海藻硫酸多糖均一性较高,通过免疫和抗氧化调节机制起一定的抗肿瘤作用。Objective To isolate and purify sulfated polysaccharide from laminaria japonica,observeits inhibition on S180 tumor,and to study the mechanism of anti-tumor activity at the level of immunoloregulation functions and antioxidant effects.Methods Sulfated polysaccharide from laminaria japonica was obtained from laminaria japonica through isolation and purification,whose uniformity was studied on Sephacryl S200 column and the monosaccharide composition of SP was analyzed by gas chromatography.The mouse model bearing S180 tumor was established,and mice were cured with sulfated polysaccharide from laminaria japonica through I.P.The inhibitory effect on S180 tumor,spleen index and thymus index were observed.NK activity and proliferation of T lymphocytes as well as antioxidant enzyme activity and MDA content of liver suspension and blood serum was detected at the same time.Results There was a single peak of sulfated polysaccharide from laminaria japonica on Sephacryl S200 column.The inhibitory rate of the low,medium and high dose of sulfated polysaccharide from laminaria japonica was 4.89%,26.63% and 29.35% respectively.sulfated polysaccharide from laminaria japonica was capable of anti-oxidation and immunoloregulation as well.Conclusion The uniformity of sulfated polysaccharide from laminaria japonica is improved after purification.Through anti-oxidation and immunoloregulation,sulfated polysaccharide from laminaria japonica can partly inhibit S180 tumor in vivo of mice.
