摘 要:目的初步建立水芹总酚酸胶囊的质量控制标准。方法采用薄层色谱法对水芹总酚酸进行定性鉴别;采用Folin酚法测定水芹总酚酸含量;采用高效液相色谱法测定总酚酸中绿原酸含量,色谱条件:DiamonsilC18柱,流动相:A:乙腈-甲醇(10∶1);B:0.4%磷酸,梯度洗脱;检测波长:325nm;流速:1ml/min;柱温:25℃。结果按现有生产工艺,水芹总酚酸胶囊总酚酸含量最高为32.42%,绿原酸最高为9.3%。结论 Folin酚法和高效液相色谱法能精确测定水芹总酚酸胶囊中总酚酸和绿原酸含量,可作为水芹总酚酸胶囊的质量控制方法 。Objective To establish a quality standard for the capsule of total phenolic acid extracted from Oenanthe Javanica.Methods Oenanthe Javanica was identified by thin layer chromatography.Folin-Phenol method was applied to the determination of total phenolic acid while high performance liquid chromatography-DAD was established to determine the concentration of chlorogenic acid in total phenolic acid.Chromatographic conditions:Diamonsil C18 column was used as the stationary phase,gradient elution was adopted with A:acetonitrile-methanol(10∶1)B:0.4% phosphoric acid.The detection wavelength was at 325 nm.Results Under the existing technology,the highest concentration of total phenolic acid was 32.42%;the chlorogenic acid was 9.3%.Conclusion The result showed that this method is reliable,accurate and suitable for quality control of the capsule of total phenolic acid from Oenanthe Javanica.
