摘 要:目的:建立盐酸安非他酮血药浓度测定方法.方法:反相高效液相色谱法,采用色谱柱:Kromasil C18(5μm,4.6mm×250mm) 流动相:甲醇:40mmol/L NH4AC[(乙酸调pH(4.5±0.1)]:(60:47,v/v) 流量:1.0ml/min 柱温:室温 检测波长:254nm 进样量:20μl.内标物:盐酸普萘洛尔.结果:以盐酸安非他酮与内标物峰面积之比(y)对盐酸安非他酮血清浓度(C)进行线性回归,得回归方程:C=1.13×102y-1.12×10,(r=0.9995,n=7),结果表明:盐酸安非他酮在5.0~500.0μg/L范围内线性关系良好.结论:本方法操作简便、回收率高、精密度好.可以用于盐酸安非他酮的治疗药物监测.Objective:To develop an HPLC method for the quantitative determination of Bupropion Hydrochloride in patient's serum. Methods: The residues were analyzed with an HPLC system Kromasil C18(5μm,4.6mm×250mm)with the mobile phase consisted of methanol: 40mmol/L NH4AC(With acetic acid adjust to pH 4.5±0.1)(60:47,v/v),The flow rate was 1.0 ml/min and the detection wavelength was at 254nm and the Column temperature was room temperature. Propranolol hydrochloride was used as internal standard.Results:Linear regression analysis was made by Bupropion hydroehloride serum concentration(C) and the peak area ratio(y) of Bupropion hydrochloride vs. internal standard(Propranolol hydrochloride),the regression equation was C=1.13×10^2y-1.12×10,(r =0.9995,n=7). The results showed that: Bupropion Hydrochloride has a good linear relationship in the range of 5.0-500.0μg/L. Conclusions:The method provides a simple,high recovery and good precision way for monitoring serum level of Bupropion Hydrochloride, and this method can be used for therapeutic drug monitoring of Bupropion Hydrochloride.
