摘 要:建立了微流控芯片非接触式电导法快速测定药物中盐酸异丙嗪含量的分析方法。研究了缓冲液、添加剂、分离电压和进样时间等因素对检测的影响,以20 mmol.L-1MES-20 mmol.L-1H is的缓冲液为分离介质,分离电压为2.2 kV时可实现较好的分离检测。在优化实验条件下,盐酸异丙嗪的线性范围为100~1 000 mg.L-1;检出限(S/N=3)为10 mg.L-1;线性方程为Y=-502+4.99×103X,相关系数r=0.995 6;0.1 g.L-1盐酸异丙嗪的RSD为2.1%(n=5);3批药品的加标回收率为96%~101%,RSD为1.7%~3.0%。该方法简便、快速、灵敏度高,可以用于盐酸异丙嗪药物的质量控制。A rapid and simple method was established for the determination of promethazine hydrochloride in injection and tablet by microfluidic chip with contactless conductivity detection.The experimental factors,including type and concentration of buffer and additive,separation voltage and sample injection time were optimized.In the optimal conditions,using 20 mmol·L-1 MES-20 mmol·L-1 His as buffer solution,separation voltage of 2.2 kV and injection time of 10 s,promethazine hydrochloride in the samples was separated and detected in 2 min.The linear relationship for promethazine hydrochloride ranged from 100 mg·L-1 to 1 000 mg·L-1.The linear equation was Y=-502+4.99×103 X with a correlative coefficient of 0.995 6 and a detection limit(S/N=3) for 10 mg·L-1.The relative standard deviation(RSD,n=5) for 0.1 g·L-1 promethazine hydrochloride was 2.1%.The proposed method was successfully applied in the detection of promethazine hydrochloride in three samples.The spiked recoveries of promethazine hydrochloride from real samples at three concentration levels of 100,300 and 500 mg/L were in the range of 96%-101% with RSDs of 1.7%-3.0%.The method could be used in the quality control of promethazine hydrochloride drug with its simplicity,sensitivity and speediness.
