免疫抑制剂FK506对乳腺肿瘤细胞株的体外作用

摘　　要：利用FK506(Tacrolimus)对乳腺肿瘤细胞株MCF-7进行体外作用研究,发现FK506可以抑制其增殖。MTT检测结果显示,24 h与48 h的半抑制浓度基本一致,说明其在较短时间内即可发挥有效作用。细胞形态学观察以及细胞染色表明,FK506没有诱导细胞凋亡,而是阻抑了细胞的分裂。进一步经双向电泳(2-DE)蛋白质表达图谱和MALDI-TOF-TOF质谱分析,发现有17种蛋白在FK506处理细胞前后发生明显的表达量上的差异,并进一步从中鉴定出12种蛋白。这些蛋白,按其功能主要分为两大类:一类参与调节细胞骨架的装配与运输,从而影响到神经发育与细胞分裂;另一类,与基因存在密切的相互作用,参与了其复制、修复、转录调节以及翻译后蛋白的折叠,从而影响细胞分裂周期与凋亡。这些差异蛋白为明确FK506抑制细胞增殖的作用机制提供了基础,同时,FK506可以抑制乳腺肿瘤细胞增殖这一功能,也为其成为临床治癌药物提供了可能性。The study finds that FK506（Tacrolimus）prevents the proliferation of breast tumor cells MCF7 in vitro;MTT assy results show 24h and 48h are basically the same on half-inhibitory concentration,indicating it can play an effective role in a relatively short period of time.Through the observation of cell morphology and cell staining,the authors detect FK506 do not induce apoptosis,but possibly inhibition of cell division.By the two-dimensional electrophoresis（2-DE）,protein expression spectra and MALDI-TOF-TOF mass spectrometry analysis,there are 17 proteins having the apparent differences in cells treated with FK506 before and after in expression levels,and further from which the authors identifiy 12 proteins.The function of these proteins can be divided into two aspects： on the one hand,regulating cell skeleton assembly and transport thus affecting the neurological development and cell division;on the other hand,there is close interaction with the gene,involved in its replication,repair,transcription regulation and post-translational protein folding,thereby affecting the cell division cycle and apoptosis.These proteins provide the basis for elucidating the mechanism which FK506 can inhibit cell proliferation,and the truth which FK506 could arrest the proliferation of breast cancer cells applies for its possibility to use as clinical cancer drugs.
